Yeast oxidative stress response

چکیده
Aerobic organisms are constantly exposed to reactive oxygen species (ROS), generated as normal metabolism byproducts, especially by respiration [1,2]. To modulate ROS concentrations and to counteract their deleterious effects, there are several antioxidant systems with an apparent functional redundancy, which may provide an evolutionary advantage. Saccharomyces cerevisiae cells possess multiple H2O2 detoxifying enzymes, such as catalases, cytochrome c peroxidase, glutathione peroxidases, glutaredoxins and peroxiredoxins, described as many isoforms and in distinct cellular compartments [3–6]. So far, their specific roles

Multistep phosphorelay proteins transmit oxidative stress signals to the fission yeast stress-activated protein kinase.
In response to oxidative stress, eukaryotic cells induce transcription of genes required for detoxification of oxidants. Here we present evidence that oxidative stress stimuli are transmitted by a multistep phosphorelay system to the Spc1/Sty1 stress-activated protein kinase in the fission yeast Schizosaccharomyces pombe. The fission yeast mpr1(+) gene encodes a novel protein with a histidine-c...
متن کاملSugar metabolism, redox balance and oxidative stress response in the respiratory yeast Kluyveromyces lactis
A lot of studies have been carried out on Saccharomyces cerevisiae, an yeast with a predominant fermentative metabolism under aerobic conditions, which allows exploring the complex response induced by oxidative stress. S. cerevisiae is considered a eukaryote model for these studies. We propose Kluyveromyces lactis as a good alternative model to analyse variants in the oxidative stress response,...
متن کاملConserved actin cysteine residues are oxidative stress sensors that can regulate cell death in yeast.
Actin's functional complexity makes it a likely target of oxidative stress but also places it in a prime position to coordinate the response to oxidative stress. We have previously shown that the NADPH oxidoreductase Oye2p protects the actin cytoskeleton from oxidative stress. Here we demonstrate that the physiological consequence of actin oxidation is to accelerate cell death in yeast. Loss of...
متن کاملProteomic Research Reveals the Stress Response and Detoxification of Yeast to Combined Inhibitors
The tolerant mechanism of yeast to the combination of three inhibitors (furfural, phenol and acetic acid) was investigated using 2-DE combined with MALDI-TOF/TOF-MS. The stress response and detoxification related proteins (e.g., Ahp1p, Hsp26p) were expressed higher in the tolerant yeast than in the parental yeast. The expressions of most nitrogen metabolism related proteins (e.g. Gdh1p, Met1p) ...
متن کاملDeteriorated Stress Response in Stationary-Phase Yeast: Sir2 and Yap1 Are Essential for Hsf1 Activation by Heat Shock and Oxidative Stress, Respectively
Stationary-phase cultures have been used as an important model of aging, a complex process involving multiple pathways and signaling networks. However, the molecular processes underlying stress response of non-dividing cells are poorly understood, although deteriorated stress response is one of the hallmarks of aging. The budding yeast Saccharomyces cerevisiae is a valuable model organism to st...
متن کاملKluyveromyces lactis: A Suitable Yeast Model to Study Cellular Defense Mechanisms against Hypoxia-Induced Oxidative Stress
Studies about hypoxia-induced oxidative stress in human health disorders take advantage from the use of unicellular eukaryote models. A widely extended model is the fermentative yeast Saccharomyces cerevisiae. In this paper, we describe an overview of the molecular mechanisms induced by a decrease in oxygen availability and their interrelationship with the oxidative stress response in yeast. We...
